马铃薯GA含量相关基因的特征分析及其在块茎形成中受BR直接调节的初步机制分析
摘 要
马铃薯( Solanum tuberosum L.) 是世界第四大粮食作物,块茎是其产量构成的基础,而赤霉素(gibberellin,GA)是最重要的块茎形成调控因素,可以抑制块茎形成。GA含量主要受到其合成、代谢和转运的影响。为了解析GA在抑制块茎形成中的作用,我们对马铃薯中已知的六个GA含量相关基因进行了特征分析。我们首先分析了它们的基因结构,发现这些基因大多数含有内含子;之后我们对这些基因分别进行了系统进化分析,发现StGA2ox1和StGA20ox1与拟南芥中对应基因的亲缘关系较近;我们还对其表达进行了组织特异性以及植物激素和胁迫处理的调控分析,发现这些基因的表达都存在组织特异性,并且适当浓度的IAA、ABA、GA3以及盐胁迫和 *51今日免费论文网|www.jxszl.com +Q: ¥351916072¥

CHARACTERISTIC ANALYSIS OF POTATO GA CONTENTRELATED GENES AND PRELIMINARY MECHANISM ANALYSIS OF THEIR DIRECT REGULATION BY BRASSINOSTEROIDS IN TUBERIZATION
ABSTRACT
Potato (Solanum tuberosum L.) is the fourth largest food crop in the world, whose tubers are the basis of its yield composition. Gibberellin (GA) is the most important inhibitor for tuberization, whosecontent is mainly affected by its synthesis, metabolism and transport. In order to reveal the role of GA in inhibiting tuberization, we conducted a characteristic analysis for six known genes related to GA content in potatoes, including gene structure, and phylogenetic trees. The results showed that most of these genes contained introns and StGA2ox1 and StGA20ox1 are close to the corresponding genes in Arabidopsis. We also analyzed the expression of these genes in different tissues and under different hormone or stress treatments.We find that the expression of all these genes are tissuespecific.The results also showed that IAA, ABA, GA3 , as well as salt stress and drought stress, have an effect on the expression of these genes. On the other hand, many studies have shown that exogenous brassinosteroids (BRs) spraying can increase the yield of potatoes in pot and field trials. In order to analyze whether the promoting effect of BR on tuberization depends on the regulation of GA contentrelated genes, promoter regions with 3000 bp upstream of these genes were collected and analyzed. We found that there is a large number of ebox and a small number of BRRE elements in the promoters, which can be bound by the BR signal transcription factor BES1 / BZR. All these results provide a theoretical basis for revealing the mechanism of direct regulation of BR on GA contentrelated genes.
