水稻半胱氨酸蛋白酶基因OsCPx调控种子萌发和幼苗生长的生理机制研究
摘要
半胱氨酸蛋白酶家族是目前研究较为广泛的一类蛋白水解酶,有研究表明其与植物体的种子萌发、幼苗生长、胁迫响应和组织衰老相关。但是水稻中半胱氨酸蛋白酶基因的功能了解的还不甚透彻。本文以日本晴野生型和oscpx突变体为实验材料,研究了OsCPx基因在水稻早期生长发育中的生理功能。基因定量结果显示OsCPx基因在种子萌发过程中表达量显著上调,表明其可能参与种子萌发和幼苗早期生长过程。与野生型相比,oscpx突变体种子萌发和幼苗生长均较迟缓,这可能主要是由于其种子内蛋白酶活性降低导致的。蛋白酶活性降低导致种子胚乳内蛋白水解能力下降,产生的氨基酸含量降低,因此影响了种子萌发和幼苗生长。这些研究结果表明OsCPx基因正向调控水稻种子萌发和幼苗生长。本研究为深入研究OsCPx基因的功能奠定了基础。

s been widely investigated recently. Some researches have shown that it is related to seed germination, seedling growth, stress response and tissue aging of plants. However, the function of caspase gene in rice has not been thoroughly understood. This research use Rice(Oryza sativa L. japonica. cv. Nipponbare) and oscpx mutants as experimental materials to explore the physiological function of oscpx gene in seed growth and early seedling growth of rice. The quantitative results shows that the expression of oscpx gene was significantly upregulated during seed germination, which indicates that oscpx gene might be involved in seed germination and early seedling growth. Compared with the wild type, the seed germination and seedling growth of oscpx mutant were stunted, which may be due to the decrease of protease activity in the seed. The decrease of protease activity results in the decrease of proteolysis ability and amino acid content in endosperm, affecting seed germination and seedling growth. These results indicate that OsCPx gene has a positive effect on seed germination and seedling growth of rice. This research laid a foundation for further study on the function of OsCPx gene.
