豌豆ALOG基因家族成员SYL7的克隆和功能研究
摘 要
ALOG(Arabidopsis LSH1 and Oryza G1)基因家族的命名来自拟南芥和水稻中两个具有代表性的转录因子LSH1和G1,序列分析发现该基因家族负责编码一类具有DUF640的保守结构域,被命名为ALOG结构域。已有的研究表明,ALOG基因家族在陆生高等植物中存在多个拷贝,编码转录因子,参与植物多个发育过程。目前,该基因家族成员在豆科植物中的功能尚不清楚。本文以豆科植物豌豆为研究材料,通过生物信息分析鉴定豌豆ALOG基因家族成员,使用MEGAX构建了豌豆ALOG基因家族的系统发育树,发现豌豆中ALOG基因可以分为三个分支,Clade 1,Clade 2和Clade 3。通过整合豌豆公共数据库和实验室本地数据,研究豌豆ALOG基因的表达模式,发现ALOG基因在不同的组织都有表达,但不同ALOG基因的表达模式差异很大。SYL7基因Psat0s2306g0040,含有3个exon和2个intron,该基因存在可变剪接。SYL7基因主要在豌豆营养组织顶端、生殖组织顶端以及花芽等组织中表达。本研究为将来进一步研究豌豆ALOG基因的功能奠定了基础。

YSIS OF ALOG FAMILY GENE SYL7 IN PEA
ABSTRACT
The gene family of ALOG (Arabidopsis LSH1 and Oryza G1) comes from two representative transcription factors LSH1 and G1 in Arabidopsis and Oryza. The ALOG family proteins have conserve domain DUF640, which is named as ALOG domain. It has been found that there are many copies of the genes in higher plants, which are involved in multiple biological processes, such as embryonic development, organ formation, flowering and fruit ripening. However, it is not clear about the expression pattern and function of the ALOG family, especially in legumes. To better characterize the ALOG family transcription factors in pea, we conducted a BLASTp search against our local database and a public database. The searches identified 17 putative ALOG proteins in pea. Phylogenetic analysis showed that these ALOG family proteins in pea could be divided into three clades. The above results suggested that ALOG family genes have undergone multiple duplication events during legume evolution. We explored the expression patterns of ALOG family genes across different tissues and organs of pea using the data from the gene expression atlas in pea and our local expression data. ALOG family genes were expressed in all tissues and organs examined, but with different levels of expression in the nodules, vegetative apices, and reproductive apices. The results showed that SYL7 gene had a segment of ALOG domain, which was mainly expressed in meristem such as vegetative shoot meristem, reproductive shoot meristem and floral buds. This study would provide a basis for further functional investigation of ALOG family genes in pea.
