中国南方大豆种质资源种子耐涝性鉴定及GmCML基因家族研究
摘 要
在我国南方地区在大豆生产的过程中,大豆植株易遭受洪涝灾害,发掘耐涝大豆种质资源和耐涝基因具有重要的理论和实践意义。本研究通过模拟淹水逆境处理种子,以相对发芽率、电导率和相对苗长为指标对我国南方的183份大豆地方品种资源进行鉴定,结果表明,供试材料种子淹水后其发芽相关性状存在较大的遗传变异,其中相对发芽率的平均值为89%,变幅为42109%;发现电导率与相对发芽率和相对苗长均存在负相关的关系;本研究以相对发芽率、电导率和相对苗长为指标,筛选出耐淹大豆地方品种资源15份。本研究同时对大豆钙调素GmCML基因家族中的两个耐涝应答基因Glyma.02G245700与Glyma.18G260700进行生物信息学分析。发现这两个基因编码的蛋白长度分别为141个氨基酸和151个氨基酸。且含有多个胁迫响应基因。通过构建系统进化树得知Glyma.02G245700与AT1G66400 c *51今日免费论文网|www.51jrft.com +Q: *351916072

IDENTIFICATION OF WATERLOGGING TOLERANCE AND GmCML GENE FAMILY OF SOYBEAN GERMPLASM RESOURCES IN SOUTH CHINA
ABSTRACT
In the process of soybean production in southern China, soybean plants are vulnerable to flood and waterlogging, so it is of great theoretical and practical significance to explore waterlogging tolerant soybean germplasm resources and waterlogging tolerant genes. Seeds treated by simulating flooding adversity, this study with relative germination percentage, electrical conductivity and the length of seedling index of southern China where 183 soybean variety resources, verification results show that the selected material seeds germinate after flooded its related traits exist large genetic variation, the relative germination rate was 89%, the average range of 42109%; It was found that there was a negative correlation between electrical conductivity, relative germination rate and relative seedling growth. Based on the relative germination rate, electrical conductivity and relative seedling length, 15 local varieties of wateredresistant soybean were selected. In this study, bioinformatics analysis was performed on two waterlogging tolerance genes in the soybean calmodulin GmCML gene family, glyma.02g245700 and glyma.18g260700. The two genes were found to encode proteins of 141 and 151 amino acids, respectively. It also contains multiple stress response genes. According to the phyletic tree, glyma.02g245700 is closely related to at1g66400calmodulin like 23. Glyma.02g245700 and glyma.18g260700 belong to the same subgroup, but belong to different groups. The results can provide reference for the genetic breeding of waterlogging tolerance of soybean.
