zebularine诱导的小麦中国春和辉县红染色体稳定性分析
摘 要
DNA甲基转移酶抑制剂zebularine由于具有稳定性高、毒性低等优点,在植物染色体工程和诱变育种等研究方面逐渐得到关注。本实验利用zebularine注射孕穗期的中国春和辉县红小麦,获得M1及M2种子,利用寡核苷酸探针FISH技术,探讨zebularine对中国春小麦和辉县红小麦染色体的诱变效应。首先构建了辉县红和中国春核型,可以清晰识别全部染色体,同时未发现存在染色体变异;其次,从45株辉县红M1植株中,发现3株存在明显变异的染色体,包括缺失del4B和端体6BS、4AL和3BL,但在22株中国春M1中,未发现明显的变异染色体,说明zebularine可以诱导小麦染色体变异,但在不同基因型间效率不同。对36株辉县红和34株中国春M2植株鉴定,尚未发现明显的结构变异,需要进一步扩大群体,验证zebularine诱导后M2代染色体变异特点。

s gradually favored by breeders in plant chromosome engineering and mutation breeding due to its high stability and low toxicity. In this experiment, M1 and M2 plants of wheat Chinese Spring and Hui xianhong induced by zebularine via booting injection were identified by oligonucleotide probe set FISH, its mutagenic effect on chromosome stability was evaluated. High resolution karyotypes of Huixianhong and Chinese Spring were devloped first and then used for identifying chromosomes of zebularineinduced plants. Among 45 M1 plants of Huixianhong, three plants involved chromosome aberrations including deletion del4B, telosomes 6BS, 4AL and 3BL, but in 22 Chinese Spring M1 plants, no aberration was found. This indicated that zebularine could induce chromosome instability of wheat via booting injection, but its effect might depend on wheat genotypes. However, no chromosome aberrations were identified from 36 Huixianhong and 22 Chinese Spring M2 plants. More sample plants need to be checked in order to verify the effect of zebularine in M2 plants of wheat.

引言:小麦是世界上重要的粮食作物,我国小麦产量与种植面积都位居世界首列。但是,由于我国人口基数大、人均耕地面积小,使得提高产量的方法只能从提高单产方面入手。然而,我国小麦育种仍然存在一些问题,例如育种单位多、组织形式落后、规模小、材料与信息交流不畅、缺乏有效合作,最关键的是主产地区所使用的亲本较为单一,从而导致新品种在遗传相似性上增加,最终导致新品种的育成难度加大(何中虎等,2011)。
