转基因抗虫棉花28025105的环境安全评价
摘 要
转基因抗虫棉花的环境安全评价是申请生物安全证书的必要环节,花粉活力和对土壤微生物群落多样性的影响是环境安全性评价的重要内容。本文针对新的转Cry2Ab4基因抗虫棉花28025105,比较了其与受体棉花中棉24的花粉活力以及对棉田微生物群落的影响之间的差异。在盛花期采集转基因抗虫棉花28025105及其对照受体材料中棉24的花朵,采用花粉离体萌发测定法测定花粉量、花粉大小以及萌发率,结果表明转基因抗虫棉花28025105的花粉数量与中棉24相比多22%;这两种类型棉花花粉大小和萌发率均无显著差异。在棉花生长的苗期、蕾期、花铃期、吐絮期和收获后期采集土壤样品,采用土壤稀释涂布平板法进行土壤微生物群落数量的测定,结果表明这两种棉田中的土壤微生物(细菌、真菌以及放线菌)在不同的时期群落数量变化规律基本一致,且同时期两者之间的土壤微生物群落数量无明显差异。上述结果表明转基因抗虫棉花28025105和其对照材料在花粉活力及对土壤微生物群落的影响方面没有明显差异,为该新转化体材料研发申请进入下一个阶段提供了重要的参考数据。 *51今日免费论文网|www.51jrft.com +Q: &351916072&
EVALUATION OF ENVIRONMENTAL IMPACT OF TRANSGENIC INSECTRESISTANT COTTON 28025105
ABSTRACT
The evaluation of environmental impact transgenic insectresistant cotton is a necessary step for the application of biosafety certificate. Its pollen vitality and potential impact on the diversity of soil microbial community are important contents of evaluation of environmental impact. In this paper, the insectresistant transgenic cotton 28025105 with new Bt gene Cry2Ab4 and its receptor cotton ZM24 were used as research materials to compare their difference on pollen vitality and microbial community in cotton fields. The flowers of 28025105 and ZM24 during the cotton blooming period were collected. The amount, size and germination rate of pollen were determined by the in vitro germination assay. The results showed that the amount of pollen of transgenic insectresistant cotton 28025105 was 22% more than that of ZM24, the pollen size and germination rate of these two cottons were not significantly different. Soil samples were collected at the seedling stage, bud stage, flower boll stage, flocking stage and late harvest stage of cotton growth, and the number of soil microbial communities was determined by the soil dilution coating plate method. The results indicated that the number of soil microorganisms (bacteria, fungi and actinomycetes) in these two cotton fields had the same pattern in different periods and there was no significant difference in the number of soil microbial communities between transgenic insectresistant cotton and the receptor cotton fields in the same period. Here it provides important reference data for the development of the new insectresistant transgenic cotton 28025105.
