摘要
采用一种新的检验思路用于检测梨火疫病菌(Erwinia amylovora),为梨火疫病菌的新型快速检测提供了新的方法。用胶体金标记梨火疫病单克隆抗体,在其中加入不同量的HRP使得McAbHRP为设置好的各种比例。利用现已掌握的技术将标记好的胶体金制备大量用于实验的免疫层析试纸条,将配置好的阴性及阳性检测液置于试纸条使其反应,剪取阴性及阳性反应T线,用电化学检测仪器分别检测其电化学信号强弱。计算同一组数据的阳性反应线与阴性反应线的电化学信号的比值,即可表示各种不同比例下的电化学检测灵敏度。多次重复实验分析比较后可得出利于梨火疫病菌快速检测的McAbHRP的最优比例。该实验合成Mc *51今日免费论文网|www.51jrft.com +Q: ¥351916072¥ 
AbAuHRP复合物作为放大信号探针,和传统的以胶体金作为主要指示物的免疫层析检测方法相比,在保证其特异性的同时还可以放大待测物的电化学信号,有效提高胶体金免疫层析方法的检测灵敏度,在梨火疫病菌的快速检测领域具有良好的应用前景。
SYNTHESIS AND CHARACTERIZATION OF THE MuAbAuHRP SIGNAL PRODE THAT IS USED FOR ERWINIA AMYLOVORA IMMUNOCHROMAROGRAPHY ELECTROCHEMICAL DETECTION
ABSTRACE
A new method for the detection of pathogen of Erwinia amylovora, it provides a new method for the rapid detection of pear fire blight. Labeling monoclonal antibodies against pear fire blight with colloidal gold and the addition of different amounts of HRP makes McAbHRP be a set ratio.A large number of immunochromatographic test strips for experiments were prepared by using the colloidal gold detection technology. Put different ratio of McAbHRP solution on the test strip to make it react and then cut the negative and positive Tline. Detection of electrochemical signal strength with electrochemical detector, Calculate the ratio of the electrochemical signal of the positive reaction line and the negative reaction line of the same set of data and the sensitivity of electrochemical detection can be expressed under various ratios. Then we can obtain the optimal ratio of McAbHRP for detection after repeated analysis and comparison. In this experiment, McAbAuHRP complex was synthesized as an amplification signal probe. Compared with the traditional immunochromatographic method using colloidal gold as the main indicator, it can not only ensure its specificity, but also amplify the electrochemical signal of the object to be tested. The signal prode can improve the sensitivity of immunochromatography with colloidal gold. So this method has a good application prospect in the rapid detection of pear fire blight.
