选高效的黄酮类化和物作为杀菌剂的主要活性成分。对烟草疫霉采取菌丝生长法,分别测定柚皮素、槲皮素、橙皮素、山萘酚、甘草素、根皮素、芦丁对其的抑菌活性。结果发现柚皮素、根皮素、橙皮素和甘草素对烟草疫霉具有良好的抑制作用,其中效果最好的柚皮素EC50值为20.19 mg/L,另外三种物质EC50值分别为30.92 mg/L, 34.91 mg/L和48.93 mg/L。同时发现,柚皮素对辣椒疫霉、终极腐霉和瓜果腐霉也具有一定的抑制作用,EC50值分别为44.12 mg/L, 76.82 mg/L和88.73 mg/L。烟草疫霉在柚皮素EC50值下液体培养基中培养48h后进行转录组测序,发现658个基因表达量上调,612个基因表达量下降,受柚皮素影响烟草疫霉在转录组水平上有所变化。柚皮素对烟草疫霉的繁殖也具有一定的影响,孢子囊萌发的EC50值为2.574 mg/L,休眠孢子萌发的EC50值为10.51 mg/L。研究结果表明柚皮素具有开发为防治烟草黑胫病的新型植物源杀菌剂的潜力。
STUDY ON A PLANT SECONDARY METABOLITES FOR THE PREVENTION AND TREATMENT OF TOBACCO BLACK SHANK DISEASE
ABSTRACT
In order to explore new measures for disease control in "green agriculture", effective flavonoids were selected from plant secondary metabolites as the main active components of fungicides. The mycelial growth of Phytophthora nicotianae was studied to determine the antimicrobial activity of naringenin, quercetin, hesperidin, kaempferol, liquiritigenin, phloretin and rutinum. The results showed that naringenin, phloretin, hesperidin and liquiritigenin had a good inhibitory effect on Phytophthora nicotianae. The EC50 value of naringenin of the most effective was 20.19 mg/L, and the EC50 value of the other three substances was 30.92 mg/L, 34.91 mg/L and 48.93 mg/L, respectively. At the same time, it was found that the naringenin also had a certain inhibitory effect on Phytophthora capsici, Pythium ultimum, and Pythium aphanidermatum. The EC50 values were 44.12 mg/L, 76.82 mg/L and 88.73 mg/L, respectively. Transcriptome sequencing of Phytophthora nicotianae was performed after 48h of culture in V8 liquid medium with the value of EC50 of naringenin. It was found that the expression levels of 658 genes were upregulated, and the expression levels of 612 genes were decreased. The EC50 value of sporangium germination was 2.574 mg/L, and that of dormant spore germination was 10.51 mg/L. The results show that naringenin has the potential to be developed as a new botanical fungicide for the prevention and treatment of tobacco black shank disease.
