萝卜(Raphanus sativus L)是原产于我国的重要根菜类蔬菜作物,因其营养丰富,并且具备一定的经济价值,所以深受人们的喜爱并在亚洲乃至世界范围内广泛种植。近年来,随着经济的发展,全球温度也在节节攀升,而萝卜是喜冷凉蔬菜作物,大多数品种适宜秋冬栽培,高温往往会影响萝卜夏季的生产和供应。因此,阐明萝卜的抗热性遗传生理机制,培育抗热性强的优良品种已成为亟待解决的问题。已知植物HSP20是一类分子量较小在生 *51今日免费论文网|www.51jrft.com +Q: *351916072*
物受到不良环境因素影响诱导合成的一类蛋白质——热激蛋白(heat shock protein,Hsps),广泛存在于从细菌到人等生物体中。在高等植物中,Hsp基因参与许多发育过程并对非生物胁迫做出响应。本研究基于16年发表的萝卜基因组,共鉴定出61个RsHSP20家族候选基因,这61个RsHSP20中有54个被定位到R1R9染色体上,RsHSP20蛋白的分子量较小且RsHSP20基因含有大量的非生物胁迫应答反应相关作用元件,在萝卜的发育过程中对非生物胁迫做出响应,而通过RNASeq数据分析显示,在萝卜不同的发育时期以及不同组织部位中有部分RsHSP20家族基因的表达具有明显的时空特异性。这些研究结果为鉴定萝卜HSP20基因的进化及其功能奠定了基础。
IDENTIFICATION OF HSP20 FAMILY GENES AND ANALYSIS OF EXPRESSION OF RADISH
ABSTRACT
Radish (Raphanus sativus L) is an important root vegetable crop originating in China. Because of its nutrientrich and economic value, it is popular among people and widely cultivated in Asia and around the world. In recent years, with the development of the economy, the global temperature is also rising and the frequency of extreme high temperature weather is gradually increasing at the same time. Radish is a cold root vegetable crop, most of which are suitable for autumn and winter cultivation. High temperature is the main factor affecting the production and supply of radish in summer. Therefore, clarifying the genetic mechanism and cultivating excellent varieties with strong heat resistance have become an urgent problem to be solved. It is known that plant HSP20 is a kind of heat shock protein (Hsps) with a small molecular weight,it is synthesized under the adverse influence of environmental factors,and it is widely found in organisms from bacteria to humans. In higher plants, the Hsp gene is involved in many developmental processes and responds to abiotic stress. This study based on the published Radish genome of 2016, 61 RsHSP20 candidate genes in the family were identified, and of these 61 RsHSP20,54 is positioned to R1 R9 on chromosome. RsHSP20 protein has smaller molecular weight and RsHSP20 gene contains a lot of elements related to abiotic stress response, these elements will make responses to abiotic stress in the process of the development of radish , and through the RNA Seq data analysis, it showed that in different growth periods and different tissue of radish, the expression of some RsHSP20 family gene has obvious specificity in time and space. These results laid a foundation for the identification of the evolution and function of the HSP20 gene in radish.
