团头鲂叉头状转录因子(foxO1)基因序列特征及表达分析
摘 要
FoxO1(forkhead box transcription factor O1)是一种能量代谢的重要调节因子。本研究通过cDNA末端快速扩增(RACE)技术成功克隆出团头鲂(Megalobrama amblycephala)foxO1a mRNA全长序列,应用生物信息学方法对其序列特征进行解析;采用荧光定量PCR技术分析了团头鲂foxO1a的组织分布图谱及高浓度葡萄糖(3g/kg,体重)灌喂条件下团头鲂foxO1a在肝脏中的表达变化。研究结果表明,foxO1开放阅读框(open reading frame, ORF)为1965 bp,编码654个氨基酸;团头鲂foxO1氨基酸序列与草鱼(Ctenopharyngodon idella)的FoxO1氨基酸序列同源性最高,foxO1在肝脏中的表达量最高,其次是肠道和脑,在脾脏和肾脏中的表达峰度较低。不同糖水平(0%, 17%, 34%和51%)日粮对肾和肠中foxO1的表达水平有显著影响;肾foxO1a表达水 *51今日免费论文网|www.51jrft.com +Q: ^351916072

GENE SEQUENCE CHARACTERISTICS AND EXPRESSION ANALYSIS OF THE HEADFORK TRANSCRIPTION FACTOR (FOXO1)
ABSTRACT
FoxO1 is an important regulator of energy metabolism. In this study, FoxO1a of Megalobrama amblycephala was successfully cloned by race The tissue distribution map of FoxO1a and the expression of FoxO1a in the liver of Megalobrama amblycephala were analyzed by fluorescence quantitative PCR. The results showed that the open reading frame (ORF) of FoxO1 was 1965 BP, encoding 654 amino acids; the FoxO1 amino acid sequence of Megalobrama amblycephala had the highest homology with that of Ctenopharyngodon idella, the highest expression of FoxO1 in liver, the second in intestine and brain, and the lowest expression peak in spleen and kidney. Different dietary sugar levels (0%, 17%, 34% and 51%) had significant effects on the expression of FoxO1 in kidney and intestine; the expression level of FoxO1a in kidney increased with the increase of dietary sugar level; the expression level of FoxO1 in intestine changed in an inverted "V" shape with the increase of dietary sugar level, in which the expression level of FoxO1 in 17% group was the lowest and that in 51% group was the highest. The expression level of FoxO1a in the liver of Megalobrama amblycephala was significantly increased 1 4 hours after oral administration of high concentration glucose. This study further enriched the basic data of FoxO1 in fish research, and provided a theoretical basis for the regulation of fish sugar metabolism.
