鸭源肠道沙门菌的分离鉴定及其生物学特性分析
摘要
沙门菌作为一种对人类和动物危害严重的食源性病原,具有重要的公共卫生意义。同时,沙门菌病的暴发也对养殖业造成巨大的经济损失。本课题采集山东省曹县某规模化养殖场的病死鸭,从编号11的病死鸭肝脏中分离细菌,采用传统的细菌学鉴定和分子生物学鉴定手段,基于分离菌的菌落形态和菌体形态初步鉴定该分离株,再通过16S rRNA测序进一步确定该细菌菌属。通过小鼠动物实验,测定该分离菌株的半数致死量(LD50),并分析其致病性;采用KirbyBauer纸片扩散法检测该分离菌株对13种抗生素的耐药情况。该分离菌株最后鉴定为肠道沙门菌(Salmonella enterica),命名为SdC11G,LD50为3.162×107CFU/mL;药敏试验显示SdC11G菌株对氨苄西林、阿莫西林、林可霉素和链霉素4种抗生素耐药。本研究初步分析了肠道沙门菌SdC11G的致病性和耐药性,确定了该养殖场大规模鸭病暴发的原因,为科学合理用药提供理论指导,也为后续开展肠道沙门菌致病机制和防 *51今日免费论文网|www.51jrft.com +Q: *351916072*

ISOLATION, IDENTIFICATION AND BIOLOGICAL CHARACTERISTICS OF SALMONELLA ENTERICA FROM DUCK
ABSTRACT
As a food borne pathogen, Salmonella has important public health significance. The outbreak of Salmonellosis also caused huge economic losses to the breeding industry. In this study, one strain of bacterium was isolated from the liver of No. 11 dead duck in a largescale breeding farm in Caoxian County, Shandong Province. By means of bacteriological identification and molecular biological identification, the bacteria were preliminarily identified by colony morphology and bacterial morphology, followed by 16S rRNA gene sequencing. The LD50 of the isolated strain was determined and its pathogenicity was analyzed by mouse experiment. Kirby Bauer disk diffusion method was used to detect the resistance of the isolated strain to 13 antibiotics and analyze its resistance. The strain was finally identified as Salmonella enterica, named SdC11G. The LD50 value was 3.162 × 107CFU/mL. The drug sensitivity test showed that SdC11G was resistant to ampicillin, amoxicillin, lincomycin and streptomycin. Our study investigated the causes of the largescale disease outbreak in the duck farm, which provides a theoretical guidance for scientific and rational drug use. Also, this study provides a material basis for the followup development of pathogenic mechanism and prevention and control measures of Salmonella infection.
