摘 要
本试验旨在探索探讨G蛋白偶联受体40 (G proteincoupled receptor 40,GPR40)和G蛋白偶联受体41 (G proteincoupled receptor 41,GPR41)这两种脂肪酸受体在物种间的同源性以及在猪不同部位的分布情况。通过NCBI下载己有的各物种的 GPR40 和 GPR41 基因序列,用MGEA软件计算遗传距离和制作遗传进化树,对 GPR40 和 GPR41 进行物种间同源性分析,并通过相对定量实时荧光PCR(Relative quantitative realtime PCR)法检测 GPR40 和 GPR41 基因在两月龄的三元杂(杜×长×大)仔猪各组织中的表达量。结果显示:小鼠和猫 GPR40 的亲缘关系最远,大鼠和小鼠 *51今日免费论文网|www.51jrft.com +Q: @351916072@
GPR40 的亲缘关系最近,牛和猪有较近的亲缘关系;大鼠和鸡 GPR41 的亲缘关系最远,牛和猪 GPR41的亲缘关系最近。GPR40 基因在仔猪的十二指肠、空肠、结肠的表达分析发现,m RNA表达在结肠表达量最高,空肠次之,十二指肠表达量最低;GPR41 基因在仔猪的十二指肠、空肠、回肠、结肠的表达分析发现,m RNA表达在十二指肠的基因的表达量最高,空肠和结肠次之,回肠的表达量最低,本实验为进一步研究脂肪酸的代谢调控机理提供了实验依据,为研究GPR40 和 GPR41 基因的功能奠定了基础。
HOMOLOGY ANALYSIS OF FATTY ACID RECEPTORS GPR40 AND GPR41 BETWEEN DIFFERENT SPECIES AND THEIR EXPEASSION IN PIGS
ABSTRACT
This study was aimed to investigate the homology of fatty acid receptors G proteincoupled receptor 40 (GPR40) and G proteincoupled receptor 41 (GPR41) between different species and their expression in different tissues of pigs. Download the GPR40 and GPR41 gene sequences of every species through NCBI, use MGEA software to calculate the genetic distance and make a phylogenetic tree, and make homology analysis of GPR40 and GPR41 between different species. The genes expression of GPR40 and GPR41 in some tissues of twomonthold ternary crossbreed pigs were quantitated with relative quantitative realtime PCR.The result shows that the genetic relationship of GPR40 between mouse and cat is the farthest,while rat and mouse is the closest,cattle and pigs have a close relationship,and the genetic relationship of GPR41 between rat and chicken is the farthest, while cattle and pig is the closest;the expression analysis of GPR40 and GPR41 revealed that the m RNA of pig GPR40 gene was expressed abundantly in the colon tissue and followed by the jejunum tissue, but extremely low in the duodenum tissue while the m RNA of GPR41 gene was expressed abundantly in the colon tissue and followed by the duodenum and jejunum tissue, but extremely low in the ileum tissue.These work laid a foundation for providing experimental basis for other fatty acid metabolism and researching the regulation role on GPR40 and GPR41.
