一株粘细菌的分离鉴定及其抗菌性质的评估
摘 要
粘细菌是一类具有捕食特性的革兰氏阴性细菌,具有典型的社会性行为特征并能产生多种具有生物活性的次级代谢产物。粘细菌的捕食特性及其所产生的多种代谢产物使其在植物病害控制方面具有重要的应用潜力。本论文的目的是分离筛选具有优良抗菌特性的粘细菌,为病害控制提供优良的菌株资源。从盐城农田采集土壤样品,采用传统的粘细菌分离纯化方法,分别使用大肠杆菌和酵母来诱导土壤样品中粘细菌子实体的生长,用直接法分离纯化粘细菌,并对其进行初步的鉴定。实验通过酵母诱导法分离得到一株粘细菌,结合对菌株的形态观察、16S rDNA和fusA,lepA,gyrB三个管家基因序列的克隆鉴定结果,鉴定该菌株为原囊菌属(Archangium sp.)。选取几种常见的植物病 *51今日免费论文网|www.51jrft.com +Q: #351916072
原菌作为供试菌株对该株粘细菌做平板对峙实验,结果表明该株粘细菌对稻瘟病菌、尖孢镰刀菌等植物病害真菌均有良好的抑制效果。本论文为植物真菌病害的控制提供了生防粘细菌菌株资源。
ISOLATION AND IDENTIFICATION OF A MYXOBACTERIA AND EVALUATION OF ITS ANTIBACTERIAL PROPERTIES
ABSTRACT
Myxobacteria are a type of Gramnegative bacteria with predatory properties, have typical social behavioral characteristics and can produce a variety of biologically active secondary metabolites. The predatory properties of myxobacteria and the various metabolites produced by them have important application potential in the control of plant diseases. The purpose of this paper is to isolate and screen slime bacteria with excellent antibacterial properties and provide excellent strain resources for disease control. Soil samples were collected from Yancheng farmland, using traditional slime bacteria separation and purification methods. E. coli and yeast were used to induce the growth of slime bacteria fruit bodies in the soil sample. The direct method was used to isolate and purify slime bacteria and conduct preliminary identification. In the experiment, a slime bacterium was isolated by the yeast induction method. Combining the morphological observation of the strain, the cloning and identification results of 16S rDNA and three housekeeping gene sequences of fusA, lepA, and gyrB, the strain was identified as Archangium sp. . Several common phytopathogenic bacteria were selected as the test strains to conduct a flat confrontation experiment on this strain of slime bacteria. The results showed that this strain of slime bacteria had a good inhibitory effect on plant disease fungi such as Magnaporthe grisea and Fusarium oxysporum. This thesis provides resources for strains of biological antisticky bacteria for the control of plant fungal diseases.
