小麦簇毛麦3V(3D)异代换系的选育及抗白粉病鉴定
摘要
小麦白粉病由禾谷白粉菌小麦专化型(Blumeria graminis f.sp. Tritici)引起,其发生与流行不仅会造成小麦严重减产,而且还会影响小麦的品质。自20世纪70年代以来,由于种植密度的提高,水肥条件的改善,矮秆品种的推广,我国白粉病的的发病面积逐年增加,已经成为小麦生产上的主要病害之一。目前,抗病品种的种植和利用是防治小麦白粉病最为经济有效和环境友好的方法。从小麦近缘物种中转移抗白粉病基因到小麦遗传背景中是抗病育种的重要途径。簇毛麦(Dasypyrum villosum (L.) P. Candargy;syn. Haynaldia villosa Schur)作为小麦的野生近缘物种,是小麦遗传改良的重要三级基因库。前期研究在小麦簇毛麦杂交后代中已经选育得到含有簇毛麦3V染色体且具有白粉病抗性的株系。本研究中,我们在 *51今日免费论文网|www.51jrft.com +Q: #351916072
簇毛麦3VL和3VS的IT(Intron Targeting)标记中筛选得到3V专化的分子标记,同时通过基因组原位杂交(GISH)、荧光原位杂交(FISH)选育得到纯合的小麦簇毛麦3V(3D)异代换系203V17,并通过接种和自然发病分析了该异代换系的白粉病抗性以及条锈病和叶锈病反应。
DEVELOPMENT OF WHEATDasypyrum villosum 3V (3D) SUBSTITUTION LINE AND IDENTIFICATION OF RESISTENCE TO POWDERY MILDEW
Abstract
Wheat powdery mildew is caused by Blumeria graminis f.sp.Tritici,Bgt. Its occurrence and prevalence will not only cause serious yield reduction, but also affect the quality of wheat grain. Since the 1970s, the incidence area of powdery mildew in China has increased year by year because of the increase of planting density, the improvement of water and fertilizer conditions and the promotion of dwarf varieties, which makes wheat powdery mildew become one of the major diseases in wheat production. At present, the planting and utilization of resistant varieties is the most economical, effective and environmentally friendly way to control wheat powdery mildew. It is an important way to transfer powdery mildew resistance genes from wheat relatives to wheat genetic background. As a wild relative species of wheat, Dasypyrum villosum (L.) P. Candargy(syn. Haynaldia villosa Schur) is an important tertiary gene pool of wheat genetic improvement. In the previous study, a line containing 3V chromosome and exhibiting resistant to powdery mildew was selected from the progeny of wheat Dasypyrum villosum hybridization. In this study, we screened 3V specific molecular markers from IT(Intron Targeting) markers of 3VL and 3VS, at the same time, homozygous wheatDasypyrum villosum 3V(3D) alien substitution line 203V17 was selected by GISH and FISH, and the resistance to powdery mildew and the response to stripe rust and leaf rust were analyzed by inoculation and natural infection.
