摘要
大豆作为一种重要的粮食兼油料作物,在世界各国都占着非常重要的地位,但是各种食叶性害虫的危害严重影响了大豆的产量与品质,而野生大豆由于有抗逆性强、适应性好等优良性状,所以利用野生大豆的抗虫研究来改良栽培大豆极其重要。本研究以113份野生大豆群体和斜纹夜蛾为研究对象,对二龄期斜纹夜蛾幼虫进行室内生物鉴定,随后利用大学开发的355KSNP芯片进行GWAS分析。结果显示一共有48个Log P≥4.0的SNP位点被定位到,其中在幼虫重上定位到46个SNP位点,抗性等级定位到2个SNP位点,最后发现在19号染色体上的SNP15、SNP35这两个位点被幼虫重和抗性等级两性状重复定位。查找SNP15标记前后80kb范围内的基因,筛选出18个候选基因并与表达谱中具有表达差异的基因进行对比,发现其中具有二氢叶酸还原酶功能的Glysoja.19G01 基因在抗性材料虫诱导第三天R3d时出现了显著表达上调。所以将Glysoja.19G01作为野生大豆的潜在抗虫基因。
USING GWAS TO LOCATE WILD SOYBEAN LOCI RELATED TO LARVAL WEIGHT AND RESISTANCE GRADING TRAITS

Soybeans, as an important grain and oil crop, occupy a very important position in all countries in the world, but the harm of various leafeating pests seriously affects the output and quality of soybeans. However, wild soybeans have strong resistance, Good adaptability and other excellent traits, so it is extremely important to use wild soybeans insect resistance research to improve cultivated soybeans. In this study, 113 wild soybean populations and Spodoptera litura were used as research objects. The secondinstar Spodoptera litura larvae were subjected to indoor biological identification, and then the 355KSNP chip developed by Nanjing Agricultural University was used for GWAS analysis. The results showed that a total of 48 SNP sites with Log P ≥ 4.0 were located, including 46 SNP sites on the larval weight and 2 SNP sites on the resistance level. The two sites of SNP15 and SNP35 were repeatedly located by the two traits of larval weight and resistance level. Find genes within 80 kb before and after the SNP15 marker, screen out 18 candidate genes and compare them with genes with differential expression in the expression profile. Significant upregulation appeared on R3d at three days. Therefore, Glysoja.19G01 was used as a potential insect resistance gene for wild soybeans .
