水稻中Rloop与染色质重组热点关联分析
摘要
Rloop由DNA:RNA杂交双链和一条被置换出的DNA单链组成,作为一种特殊的染色质结构Rloop对生物体的生长发育有着多方面的影响。染色质重组是生物体创造遗传多样性的重要途径,重组的发生主要聚集在热点区域。在植物科学领域,对Rloop结构的研究还处于起步阶段,开展全基因组水平的Rloop研究,揭示其与染色质重组的关系,对研究作物中的遗传变异和其它重要生物学过程的分子机理有着重大意义。本课题利用水稻幼苗的dDRIPseq和重组热点数据,首先分析Rloop在基因组的分布特点,然后关联Rloop和重组热点并进行初步探究,发现与Rloop关联的重组热点更多出现在基因间区并且拥有更高的LRT值(sequenceLDhot likelihood ratio test),最后还对基因组内Rloop结构与重组热点的距离进行了分析。同时还使用了玉米叶片的dDRIPseq和重组热点数据进行同样的处理和分析,对得出的部分结论加以验证。
ANALYSIS OF CORRELATION BETWEEN RLOOP AND RECOMBINATION HOTSPOTS IN RICE
ABSTRACT
loop consisting of DNA:RNA hybrid strands and a single DNA strand is a special chromatin feature with many effects on development of organisms. Chromatin recombination plays an integral role in organisms to create genetic diversity, and recombination occurs in hotspots region mainly. In the field of plant science, the research on Rloop is still in its infancy. It is important to conduct genomewide research of Rloop and reveal correlation between Rloop and recombination hotspots to study genetic variation and internal molecular mechanism in crop. In this study, dDRIPseq and recombination hotspots data of rice seedlings were used to analyze the distribution features of Rloop in the genome, then the recombination hotspots and Rloop were associated to perform some preliminary analysis. We find hotspots related to Rloop are more likely to occur in the intergenic region and have higher LRT(sequenceLDhot likelihood ratio test). Finally, the distance between Rloop and recombination hotspots was analyzed. At the same time, we also used dDRIPseq and recombination hotspots data of maize leaves for the same processing and analysis to verify these conclusions.
