Preparation , Characterization and evaluation of cell cytotoxicity of silver nanoparticles

Abstract:Clinical viral disease caused a huge threat to the health of the pet , and silver nanoparticles has received much attention as a very promising antiviral drug. But, the safety of silver nanoparticles has been the focus of attention. In this study, gallonic acid was used as a reducing agent and stabilizer to synthesize nano silver and characterized by characterization. Besides, the cytotoxicity of cat kidney cells was evaluated by cell culture in vitro. The physichemical properties of AgNPs were further characterized. The image of transmission electron microscopy (TEM) showed that SNPs exhibited approximately spherical shape with the average diameter of 13.81 ± 2.21 nm. The absorbance peak of obtained SNPs was sharp with the maximum wavelength of 400.5 nm by ultravioletvisible (UVvis) spectroscopy. Their hydrodynamic diameters were 21.35 ± 0.90 nm , and zeta potentials were 38.6 ± 0.91 mV, respectively. These data suggested that AgNPs had the good dispersivity in the solution and the particle size is uniform. MTT assay showed that the survival rate of cells increased with the decrease of nanosilver concentration, and the cell viability was over 80% when the concentration of nanosilver was in the range of 0.25 ~ 8 μg / mL. Indicating that the cytotoxicity of nanosilver is very small. In summary, the silver nanoparticles prepared by the green reduction method of gallic acid have uniform particle size, high purity, good dispersibility and low cytotoxicity at low concentration to cat kidney cells, which has a certain application prospects In the veterinary clinic.
