新型除草剂Aspterric acid的提取分离
摘 要
化学除草剂的广泛使用已引发了严重的环境、健康和生态问题,而天然产物除草剂具有环境破坏力低、易降解、毒性低等优点。因此,天然产物除草剂的研究开发具有广阔前景。Aspterric acid (AA)是近期被发现的一种具有全新除草剂靶标的新型除草剂,本研究建立并优化了AA的提取分离方法。土曲霉Aspergillus terreus NIH2624经过PDA培养基培养以及ME发酵培养基发酵,发酵产物经过过滤、乙酸乙酯萃取、旋蒸后,再通过两次硅胶柱层析、三次反向柱层析与两次凝胶柱层析分离AA,通过TLC薄层色谱点板方法与标准品进行对比,合并相似段并找出AA所在分析段,将最终所获得的样品与标准品进行对比,得到纯度较高的AA。本研究可以为开发拥有全新靶标的天然产物除草剂奠定基础。此外,本文还分析并计算了最终所获AA的终产率和产量,提出了一些实验中面临的问题和解决方案,并展望了天然产物除草剂发展的机遇与挑战。

OF A NEW NATURAL PRODUCT HERBICIDE ASPERRIC ACID
ABSTRACT
The widespread use of chemical herbicides has caused serious environmental, health, and ecological problems, and natural products as herbicides have the advantages of low environmental damage, easy degradation, and low toxicity. Therefore, the research and development of natural product herbicides have broad prospects. Aspterric acid (AA) is a rescently identified natural product herbicide with a new mode of actin.
Here, we established and optimized the extraction and separation method of AA. The Aspergillus terreus NIH2624 was cultured in PDA medium and fermented in me medium, After fermentation, the fermentation products were filtered, extracted with ethyl acetate and rotated, then separated by two silica gel column chromatography, three reverse column chromatography and two gel column chromatography. The TLC thin layer chromatography was used to compare the plate with the standard product of AA. Finally, the obtained sample was compared with the standard product to obtain AA with higher purity. This study can lay a foundation for the development of natural product herbicides with new targets. In addition, this paper analyzes and calculates the final yield of obtained AA, also proposed some problems and expected feasible solutions in the experiment, and prospected the opportunities and challenges of the development of natural product herbicides.
